(2022,NBT) Systematic discovery of recombinases for efficient integration of large DNA sequences into the human genome

Large Serine Recombinases(LSRs) are DNA integrases that facilitate the site-specific integration of mobile genetic elements into bacterial genomes. This alone allows large DNA fragment insertion (>7kb) with high genome integration efficiencies(40-75%) without Double-Strain Breaks(DSB).

This apporach could be virus-free.

There are 3 kind of LSRs as described in the NBT paper: landing pad, genome-targeting and multi-targeting.
